Dale Denton (Seth Rogen), een luie wietroker, heeft maar één reden om zijn even luie dealer Saul Silver (James Franco) op te zoeken: om wiet te kopen, vooral het zeldzame 'Pineapple Express'. Als hij getuige is van een moord, die wordt gepleegd door een gewetenloze politieagent en de gevaarlijkste drugsbaron van de stad, raakt hij in paniek. Van schrik laat hij zijn 'Pineapple Express'-joint vallen. Deze is zo zeldzaam dat de moordenaars direct naar Saul en Dale worden geleid. De hallucinerende heren slaan op de vlucht en ontdekken al snel dat ze niet lijden aan achtervolgingswaanzin veroorzaakt door hun overmatige drugsgebruik...
