De film is gebaseerd op twee waargebeurde verhalen over twee vrouwen die vijftien jaar lang in een strijd verwikkeld zijn. Annie Parker wordt, net als haar moeder en zus, gediagnosticeerd met borstkanker. Ondertussen ontdekt geneticus Mary-Claire King, na vele pogingen, het BRCA1 gen en diens link met borstkanker. Een ontdekking die voorgoed een verschil zal maken.
