Op het hoogtepunt van de Koude Oorlog, begin jaren '60, worden de jarenlange vijanden CIA-agent Napoleon Solo en KGB-agent Illya Kuryakin gedwongen samen te werken. Zij moeten namelijk de strijd aanbinden met een mysterieuze, internationale misdaadorganisatie die door middel van kernwapens en technologie het wankele machtsevenwicht dreigt te verstoren.
