De jaren '30. Een groep filmmakers reist af naar een legendarisch eiland om een film op te nemen. Eenmaal aangekomen blijken alle geheimzinnige verhalen over het eiland op waarheid te berusten. Het eiland wordt bevolkt door mysterieuze inboorlingen en vele gigantische beesten. Hoofdrolspeelster Ann Darrow wordt de gevangene van één van deze beesten: de 8 meter grote gorilla Kong.
