Buster Moon is een erg enthousiaste en optimistische koala die de eigenaar is van een slecht draaiend theater. Om zijn schulden af te lossen en de lege zalen te vullen, besluit hij om een grootse zangwedstrijd te organiseren. Buster weet nog net 1000 dollar bijeen te schrapen als prijzengeld voor de wedstrijd. Door een fout van zijn assistent, een oude hagedis, staat echter op de flyers dat er 100.000 dollar te winnen is.
