Een ongeval in een medisch laboratorium geeft aan een Beagle-jachthond genaamd Shoeshine een paar bijzondere superkrachten: hij wordt hoogbegaafd en kan spreken. Het is een geheim dat de hond uiteindelijk moet delen met een 12-jarige jongen genaamd Jack (Alex Neuberger) die zijn eigenaar en vriend wordt. Shoeshines bijzondere eigenschappen komen goed van pas als hij zijn liefje 'Sweet' Polly Purebred en de burgers van Capitol City moet beschermen tegen de kwaadaardige Simon Bar Sinister (Peter Dinklage).
