De Grieks-Amerikaanse Toula Portakalos (Nia Vardalos) is duidelijk het zorgenkindje van de familie. Ze is 30 jaar, is niet getrouwd en heeft nog geen kinderen op de wereld gezet, terwijl dat toch het levensdoel van de Griekse vrouw is? Als alle bemoeienissen van haar familie haar teveel worden en ze de niet-Griekse Ian (John Corbett) ontmoet, besluit ze haar leven drastisch om te gooien. Ze ondergaat een totale metamorfose, verlaat het oude vertrouwde Griekse familierestaurant en besluit te trouwen met Ian. Binnen haar familie ontstaat hierdoor echter grote paniek...
