Dr. Sherman Klump is een vriendelijke, briljante genetica professor die nogal last heeft van overgewicht. Wanneer de mooie Carla Purty op de universiteit les komt geven, wordt Sherman prompt straalverliefd en zoekt wanhopig naar middelen om zijn 200-kilo zware lichaam terug te brengen naar algemeen geaccepteerde normen en zo haar hart te winnen. Hij neemt een slok van zijn experimentele vetverminderende serum en verandert in Buddy Love, een gladde afgetrainde Don Juan.
