Mike Lowrey en Marcus Burnett zijn partners bij de Miami politiemacht. Als er een lading drugs uit het politiebureau wordt gestolen hebben ze 72 uur om het terug te vinden. Om ervoor te zorgen dat een getuige mee wil werken moeten ze zich als elkaar voordoen en dat is niet zo makkelijk aangezien Lowrey een losbandige vrijgezel is en Burnett netjes getrouwd.
